The paper describes: (1) histotypic aggregation in vitro of dissociated cells of chick and mouse embryos, with emphasis on selective contact and grouping phenomena in intra-and interspecific cell combinations and (2) recent experiments bearing on the mechanism of histogenetic cohesion of dissociated cells. The procedure of aggregation by rotation provides a readily controllable experimental system suitable for quantitative studies on histogenetic interactions of dissociated cells. The restrictive effects of environmental (low temperatures, periodate) and cellular (age, growth in mono-layers) parameters on cohesive competence and histotypic association of cells were studied. Cell-free supernatants were obtained which promoted markedly the recohesion of dissociated cells at suboptimal temperatures. The cell-type specificity of these supernatants was examined. Evidence presented indicates that the characteristic cell-binding activity of the supernatants depends on products released by the cells. These results corroborate the previously advanced working concept that histotypic reassociation of embryonic cells may involve the function of specific extracellular or cell-surface materials. This assumption is re-examined in the perspective of recent findings and of analogous situations in other systems.
