Summary Mammary tissue and milk samples from eight lactating cows were examined to determine the effects of prolonged Corynebacterium bovis colonization on the humoral immune response. Plasma cells producing immunoglobulin (Ig)G1 and IgG2 predominated in all quarters, while IgA- and IgM-producing plasma cells were less abundant. Numbers of plasma cells were highest at the Furstenberg's rosette area followed by parenchymal and streak canal tissue areas in all quarters. Significantly more IgG1-, IgA-, and IgM-producing plasma cells were observed in parenchymal tissue colonized with C. bovis than control quarters. However, there were only slight increases in Ig concentrations in milk of C. bovis colonized quarters as than in controls. Indices of relative occurrence of lacteal Ig indicated that IgG1, IgA, and IgM were accumulated selectively, while IgG2 was most likely transferred passively from serum. Enhanced numbers of Ig-producing plasma cells in teat and tissues of C. bovis colonized quarters may be a source of resistance against attachment and colonization by more pathogenic mastitis-causing organisms.
