The respiratory effects of i.v. amphetamine were studied in decerebrate-unanesthetized and intact anesthetized rats. A consistent increase of respiratory frequency, minute volume and sighing activity was induced by amphetamine in both preparations, whereas the tidal volume was inconsistently modified. At high doses of amphetamine (6.0 mg/kg) the breathing was very fast and shallow. Amphetamine shifted the relationship between CO2 and minute ventilation to the left, but the slope remained parallel. It is proposed that amphetamine stimulates specifically the respiratory mechanisms which provide the inspiratory off-switch required to interrupt inspiration. Intracerebroventricular phentolamine (100 .mu.g), but not propranolol (100 .mu.g), inhibited the stimulatory effects of amphetamine (3 and 6 mg/kg, i.v.) on respiration, as well as the depressant effect on blood pressure. It is suggested that a central .alpha.-adrenergic mechanism mediates the action of amphetamine on the respiratory centers, as it does for the action of adrenergic drugs at the level of the vasomotor center.
