Combination regimens of cytotoxic chemotherapeutic drugs are a mainstay of cancer treatment. Historically, the integration of several agents with distinct mechanisms of action and non-overlapping toxic effects yielded curative regimens for some solid and haematological malignancies. 1 At present, the clinical development of targeted drugs that block specific oncogenic pathways is proceeding along a similar course. For example, selective BRAF inhibitors, the use of which achieves tumour regressions in most patients with malignant melanoma who harbour a V600E mutant BRAF allele, are now being assessed in conjunction with MAP2K (MEK) inhibitors in an effort to suppress the emergence of drug-resistant clones. 2
