The methylation status of the dengue-2 flavivirus intracellular 40 S RNA was examined. This RNA contained a single type I cap per molecule of the form m7G(5'')ppp(5'')Amp. Less than 1 molecule in 5 contained adenine base methylation either in the cap or internally. The [African green monkey kidney] Vero host cell poly A+ RNA contained both types I and II caps and internal N6-methyladenine. Possible implications of the differences between dengue virus methylation and that of the host RNA and other mRNA are discussed.
