The effects of high and low ligation of the anterior descending coronary artery myocardial cyclic[c]AMP levels were studied in open-chest dogs and baboons by means of sequential epicardial mini-biopsies (10 to 15 mg wet weight) within 1 h of ligation. In the ischemic zone of baboon hearts with low ligation there was a late increase in the level of cAMP which started about 10 min before the onset of ventricular fibrillation. In dogs with low ligation there was no such increase in myocardial cAMP within 1 h and no occurrence of ventricular fibrillation. There was also no rise of myocardial cAMP levels within seconds of low ligation in dogs or in baboons. However, cAMP was elevated in the ischemic zone in 50% of dogs within 1 to 3 min of high ligation but this rise was preceded by an accumulation of tissue lactate, suggesting that cAMP did not play a role in the early activation of glycolysis. The accumulation of cAMP in ischemic tissue may predispose to ventricular fibrillation.
