Gastric acid secretion results in a pH gradient of about 6.6 units across the secretory canaliculus of the mammalian parietal cell. That this depends directly on ATP and no other process can be shown in isolated, permeabilized gastric glands where, in the presence of mitochondria1 inhibition and anoxia, which collectively reduce acid secretion to zero, added ATP is able to restore secretion.'.'In these isolated glands, as in the intact stomach, secretion is also dependent on the presence of K'and C1-. With the isolation of an ATPase that was K+ stimulated and in vesicular form was able to pump protons, it would appear that the general mechanism for acid secretion by the stomach was established?-'In this paper we will discuss some of the mechanistic aspects of the gastric ATPase that have been elucidated, showing both similarities to and differences from the better studied Na+ K+-and Ca"-ATPases.
