Materials and MethodsPreparation of Membranes. Nicotinic acetylcholine receptor (nAcChoR) rich membranes were obtained from the electric organ of Torpedo nobiliana by differential and sucrose density gradient centrifugation according to procedures adapted from Cohen et al.(1972). Thebuffer used was Ca2+-free Torpedo Ringer (TR), pH 7.4. Membranes, at a total protein con-centration of 10 mg-mL'1 and specific activity of 1.2 µ of nAcChoR-(g of protein)" 1, were stored at-70 C in 0.3-mL aliquots.
