Using hepatocytes from fasted rats, we show that glucose can inhibit by nearly 50% the rate of its synthesis from the most important physiological substrate, lactate. Two factors are necessary to demonstrate an effect of this size: (i) The cells must be preincubated with glucose, and, equally as important (ii) lactate must be used at low substrate concentrations. While the ultimate mechanism of the glucose effect is not known, we find that, in terms of metabolic fluxes, glucose causes a diversion of flux from the pathwayP-enolpyruvate → glucose to the pathwayP-enolpyruvate → pyruvate, with pyruvate carboxylation remaining essentially constant. The effect appears to involve at least some aspects of a change,in vitro, from a fasted to a fed state.
