Abstract Acute and/or chronic life stressors have long been postulated to play a role in the pathogenesis of numerous psychiatric diseases and in the exacerbation of symptoms. Some of the evidence from life events research supporting a link between stress and psychiatric disease is reviewed. The evidence that in general dysregulation of the hypothalamic-pituitary-adrenal (HPA) axis, and specifically of corticotropin-releasing factor (CRF), the central mediator of the mammalian stress response, plays a pre-eminent role in the pathophysiology of mood and anxiety disorders is described.
