Five new zwitterionic hydrogen ion buffers are described for the 1st time: 3-(N-morpholino)-2-hydroxypropanesulfonic acid, 3-[N-bis(hydroxyethyl)amino]-2-hydroxypropanesulfonic acid, 3-[N-tris(hydroxymethyl)methylamino]-2-hydroxypropanesulfonic acid, N-hydroxyethylpiperazine-N''-2-hydroxypropanesulfonic acid and piperazine-N,N''-bis(2-hydroxypropanesulfonic acid)dihydrate. These buffers, all N-substituted 3-amino-2-hydroxypropanesulfonic acids, with pKa between 6.9-7.9 were subjected to a number of stringent tests in mammalian tissue culture [human embryonic lung fibroblast WI-38 cells, adult human skin fibroblast cells, human cervical carcinoma HeLa cells and Chinese hamster ovary cells], plant pathology [Chlorella pyrenoidosa growth effects and spinach chloroplast effects] and virology [cowpea mosaic virus infection of soybean protoplasts] to detect inhibitory effects. They appear to be equivalent to or better than any other buffers heretofore available.
