The weight of the cerebral cortex has been found to be significantly greater in rats stimulated by a complex environment than in their isolated littermates. Though small (5%), this difference was so consistent as to require histological investigation. Male S1 rats (University of California Psychology Colony) were used. At 25 days of age, pairs of littermates were taken, one animal of each pair being placed in one of the following groups: The first [Environmental Complexity and Training] (ECT) contained ten animals, housed together in a large cage and given wooden "toys" and maze experience. The second [Isolated Condition ] (IC) involved 10 animals isolated in individual cages. The experiment was replicated completely. At 105 days of age the rats were decapitated, and the brains placed in 10% formolTsaline. Transverse, frozen sections were cut at 25 micra. Subcortical landmarks were used to identify sections taken from the somatosensory and visual cortex. Tissues stained with Windle''s modified Nissl stain were used for cortical depth measurements and differential cell counts. On adjacent tissues stained with hematoxylin and eosin, blood vessel measurements were taken. The depth of cortex, measured from the pial surface to the corpus cal-losum with an optical micrometer, was significantly greater for ECT than IC animals in the visual area. In the somatosensory area the differences were not as consistent. Neurons, glia and capillaries were less numerous per field in the animals experiencing the enriched condition. However, there were more large vessels per field in the ECT animals.
