The cellular localization of the uptake of [3H] GABA and [3H]L-glutamic acid was studied in cultured dorsal root ganglia (DRG) using autoradiography. In the explant and dense zones of DRG cultures, where neurones are surrounded by satellite glial (SG) cells, both amino acids were only taken by glial cells but not by neurones. In contrast, isolated DRG neurones which are deprived of their glial envelopment revealed a strong accumulation of [3H]GABA and [3H]L-glutamic acid. It is suggested that SG cells form an effective barrier which prevents the uptake of GABA and glutamate into neuronal cell bodies.
