We have cloned the genomic cDNA of a pathogenic swine vesicular disease virus (SVDV) isolated in Ibaraki in Japan (Ji '73) (1) and the complete nucleotide sequence was determined. The cDNA library was composed of 5 clones, which were overlapped each other and covered the entire SVDV RNA sequence except 65 bases of 5' terminal region. The sequences of cDNA were determined in both directions using Sequenase kit (United States Biochemical Corporation, USA) and that of the 5' terminal region was determined by direct RNA sequencing. The genome is 7,401 nucleotides in length, excluding poly(A) tail. The DNA sequence homologies were 98.9% with SVDV H3'76 (2), 98.7% with SVDV UK'72 (3), 78.9% with coxsackievirus B (CB) 5 (4), 76.9% with CB1 (5), 76.6% with CB3 (6), 75.9% with CB4 (7), 74.7% with coxsackievirus A (CA) 9 (8) and 61.9% with poliovirus type 1 Mahoney (PVI) (9). To determine the phylogenetic relationships among SVDV and other related enteroviruses, we constructed evolutionary trees using neighbor-joining methods.
