Lateral segregation of photosystems I and II implies that linear electron transport is mediated by long-range electron carriers, with plastoquinone, plastoquinol and plastocyanin being obvious candidates (1,2). The extent of involvement by the latter carriers is determined in part by their distributions relative to that of the cytochrome bf complex (2). Recent evidence favors a homogeneous spatial distribution of cytochrome bf (2), but functional homogeneity remains problematical. Most evidence is inconsistent with a capacity for long-range electron transport by plastocyanin, but it has been argued that if plastoquinol is the only long-range carrier, then cytochrome bf in the grana could function only in cyclic electron transport (1). The rate-determining step of linear electron transport is the reduction of the Rieske iron-sulfur component of the cytochrome bf complex by plastoquinol, which occurs with a half-time of 10–20 ms (3,4). As for any other bimolecular reaction, the rate of plastoquinol oxidation has two theoretical limits: an upper, diffusional limit determined by the relative mobility of plastoquinol and a lower, chemical limit determined by activational barriers, e.g.
