Sequence analysis of 18S ribosomal RNA gene in Fragaria x Ananassa Duch. cultivated octoploid strawberry

A ribosomal DNA clone (Fr1) was isolated from a λ genomic library of octoploid strawberry by screening with a maize rDNA probe. This clone was restricted withBamHI andEcoRI and subcloned into plasmids. The strawberry 18S rRNA gene was located on 3 plasmids by hybridization with a wheat 18S rRNA gene probe. The 3 inserts contained overlapping sequences of the strawberry 18S rRNA gene, which was found to be 1804 bp long. Comparisons were made between this 18S rRNA gene and those from other plants, lower eukaryotes and animals, and as a result 6 highly variable regions were defined in the strawberry gene. The nucleotide sequence data have been submitted to the EMBL/GenBank/DDBJ databases under the accession number X15590.
