There has been considerable speculation for many years about the chemical composition of the myelin sheath of nerve fibers. Estimates of myelin composition can be made from indirect measurements, such as comparative analyses of gray and white studies of constituent increases in the nervous system during periods of rapid myelination,6-'o and studies of nerve degeneration.". l2 These estimates were fairly consistent and taken together with the many excellent physical studies on rnye l i r~ , '~-~~ give us our present picture of myelin as a helically-wrapped extension of a cell membrane, having alternate layers of lipid and protein, essentially a condensed system of unit rnernbrane~,~, the major portion of which is lipid. Within the last few years, several groups have devised procedures for the isolation of myelin in pure f ~ r m . * ~ ~ ~ The fmt chemical data on pure myelin is beginning to appear24-26. 31, 33. 3 4 and it should not be long before we have a considerable amount of reliable data on the composition of pure myelin. This paper will discuss our own work on myelin from bovine brain and will concentrate primarily on the composition of the lipid portion.
