A fixed, independent of body weight, load of glucose (100 mg.) given by stomach tube to obese-hyperglycemic mice(obob)caused a blood-glucose response almost identical to that of lean animals, but a glucose load proportional to body weight (5 mg./Gm.) provoked marked and sustained hyperglycemia. A weighted dose of glucose (1 mg./Gm.) given intravenously showed similar volumes of distribution and clearance rates in obese and lean animals. The apparently normal rate of glucose clearance of the obese animals is abnormally low considering the marked enlargement of their adipose tissue mass. On the basis of experiments performed on young animals (three weeks of age), it is suggested that the enlargement of adipose tissue mass in theobobprecedes the occurrence of significant elevations of blood glucose and serum insulin.
