The stability of the secondary and tertiary molecular structure of a protein is dependent upon the nature of its solvent environment, in addition to such factors as temperature, ionic strength and pH (Schachman, 1963; Straub, 1964). In medicine, dimethyl sulfoxide (DMSO) is generally used a t body temperature and in relatively high concentrations (Jacob et al., 1964; Kligman, 1965), conditions that would be expected to alter the conformation of proteins (Hamaguchi, 1964; see Rammler & Zaffaroni for summarizing references). In general, conformational changes are determined by measuring a difference in some physical property of the protein (Hermans, 1965); however, solvent-induced changes in enzymatic activity are also useful indicators of protein conformational changes, because an enzyme’s activity is generally greatly affected by changes in protein structure (Singer, 1962; Monod et al., 1963). In this preliminary report, we wish to present results of our investigations concerned with the effect of DMSO on several enzymatic systems and the recovery of these systems after removal of the DMSO.
