Erratic results in PEG [polyethylene glycol] estimation may be obtained for 1 of 2 reasons when protein is present in the PEG-containing sample. Any protein which remains in solution after attempted precipitation with TCA [trichloroacetic acid] may increase the optical density in the final reaction whether or not PEG is present. PEG tends to be coprecipitated with, or adsorbed on to, protein precipitates. Quantitative recovery of PEG is possible if the original sample [human serum] is diluted to contain not more than 3.0 mg protein/ml.
