THE RETROVIRAL VECTOR - REPLICATION CYCLE AND SAFETY CONSIDERATIONS FOR RETROVIRUS-MEDIATED GENE-THERAPY

The unique replication cycle of retroviruses makes them us& as vectors for gene therapy; most human gene therapy trials to date have used retroviral vectors.'However, in the long term two properties of retroviral replication lead to safety concerns about retrovirus-mediated gene therapy: the high rate of genetic variability and the random integration of the provirus into the host chromosome. The assessment of the balance between the benefits of retrovirusmediated gene therapy and the risk associated with infection with a retroviral vector is similar, in principle, to the assessment of the benefit-risk balance of using chemotherapeutic agents to treat cancer. We will discuss safety considerations for retrovirus-mediated gene therapy in the context of the retroviral replication cycle. Also, we will present a new approach that we are developing for using retroviral vectors to prevent diseases caused by retroviruses. The safety considerations for retroviral vector vaccines are similar to those for retrovirus-mediated gene therapy.
