Expts. materially broadening the basis for use of 17D virus in vaccinating against yellow fever are reported. These expts. make it possible to define in greater detail the conditions necessary for the achievement of satisfactory results and reaffirm the satisfactory nature of these results when the necessary conditions have been met. Substrains of 17D were found to be of closely similar antigenic character. Evidence that small virus doses might result in a better immune response than larger doses was also obtained. However, in order to immunize a satisfactory percentage of persons vaccinated, the routine vaccinating dose should contain not less than approx. 500 for mice. The immune response of individuals up to age 14 is directly related to age. The average protective capacity of the sera increases with the age of the donor. Human susceptibility to intra-dermally or intramusc. inoculated virus is greater than that to virus administered subcut.. but the differences are not sufficient to outweigh the advantages of the subcut. route when an adequate volume is employed.
