Abstract Fluorescence due to intracellular reduced pyridine nucleotide has been examined in vivo at different points at the surface of mammary tumours transplanted in a skin chamber in mice. Changes in the tumour fluorescence were registered according to whether animals were breathing oxygen at normal or at reduced concentrations. The fluorescence increase in hypoxia represents 75–100 per cent of the aerobic fluorescence at all well-vascularized points. At poorly vascularized tumour points or at those adjacent to necrotic centres the fluorescence increase in hypoxia was minimal. The results described directly corroborate the existence of hypoxic areas in mammary tumours of the mouse. These areas were probably caused by defects in the capillary circulation of the tumour.
