3 tables form the core of this presentation which describes a microtechnique for determining the presence of sperm-immobilizing factor in cervical mucus. Table 1 is a flow chart showing the procedure of the original tube method of sperm immobilization testing; table 2 shows procedures of the microsperm immobilization test which uses a disposable microchamber to examine antibody in small specimens of cervical mucus; table 3 describes the sperm preparation. 2 other tables summarize results of sperm immobilizing tests: table 4 gives 1 2 and 3 hour results for 15 patients of both the tube and micro method for comparison; table 5 presents results of the microsperm immobilizing test when using cervical mucus from women with demonstrated sperm-immobilizing antibody in serum. It was clearly shown that 2/3 cervical mucus samples possessed the sperm-immobilizing factor. Sperm immobilization values in cervical mucus from sterile women who did not have the sperm-immobilizing antibody in their serum showed negative sperm immobilization values in the cervical mucus. The positive sperm immobilization values in cervical mucus from sterile women who did not have sperm-immobilizing antibody in their serum showed results that strongly suggested that local antibody production to spermatozoa in the cervical canal could be possible. The microtechnique to prove the sperm-immobilizing antibody uses a smaller amount of cervical mucus specimen and inactivated complement instead of control negative serum used in the tube method; both titers i.e. original tube and new micro showed parallel values.
