The isolation of five strains of a new virus from the human respiratory tract is described. Four of the individuals had mild afebrile upper respiratory infections. The virus, 229E, is ether-sensitive, RNA, about 89 mu in size, and grows in human kidney and human diploid cells but not in monkey kidney or H. Ep. 2 cultures. It does not hemagglutinate guinea pig red cells. It is not neutralized by any of the following anti-sera: respiratory syncytial, measles, mumps, parainfluenza 1, 2, 3, 4, SV-5, influenza C. Paired sera of the five persons from whom this virus was isolated showed rises in neutralizing antibody titer, and all but one also had rises in CF antibody.
