(TCDD) § and structurally related chlorinated aromatic hydrocarbons, the induction of aryl hydrocarbon hydroxylase (AHH) activity:(2) to review the evidence that there is a stereospecific recognition site which mediates the induction of this enzyme activity;(3) to compare the structure activity relationship for binding to this receptor and the structure activity relationship for the toxicity of these chlorinated aromatic hydrocarbons; and (4) to suggest a model for the mechanism of the toxic action of these compounds.The toxicology of the halogenated aromatic hydrocarbons (the halogenated dibenzo-p-dioxins, dibenzofurans, azo-and azoxybenzenes, and biphenyls) is reviewed elsewhere in this symposium; however, it is useful to summarize some of these effects first so that we share a common understanding when the term toxicity is used in this report. TCDD was selected as the prototype compound, because it is the most potent and the most thoroughly studied. TCDD produces a variety of toxic effects in laboratory animals and human beings. The histopathologic responses produced by TCDD differ quantitatively and qualitatively in different species and to some extent in acute versus chronic administration.(1) Lethality. The cause of death remains unknown. There is an order of species sensitivity with guinea pigs, and chickens being very sensitive, rats intermediate, and mice showing less sensitivity. Following the
