CHARACTERIZATION OF ANTI-REOVIRUS IMMUNOGLOBULINS SECRETED BY CLONED HYBRIDOMA CELL-LINES

Nineteen hybridoma cell lines derived from the spleen cells of a mouse immunized with reovirus serotype 3 (strain Dearing) were cloned and the IgG secreted by them were characterized. Four IgG were directed against polypeptide .mu.NS and 2 against polypeptide .sigma.NS, the 2 nonstructural reovirus-specified polypeptides. Two IgG were directed against polypeptide .lambda.2 and 2 against polypeptide .sigma.2, both components of the reovirus inner capsid shell. Four IgG were directed against polypeptide .sigma.1, the most type specific of all reovirus-specified polypeptides, 3 against polypeptide .mu.l/.mu.lC and 1 against polypeptide .sigma.3, all components of the reovirus outer capsid shell. One IgG was directed against a complex composed of polypeptides .mu.l/.mu.lC and .sigma.3, but failed to react with either polypeptide alone. Thus the 19 hybridoma cell lines secreted IgG with specificities for 7 of the 10 reovirus-specified proteins. The ability of all these IgG to precipitate the proteins specified by the 2 heterologous reovirus serotypes, serotypes 1 and 2, was measured.
