The sialic acid and the amino sugar moieties of glycosphinoglipids were N-deacetylated (or N-deglycolylated) by hydrazinolysis without considerable (5%) N-deacylation of ceramide residue. The total recoveries of various glycosphingolipids after hydrazinolysis were globoside, 52%; neolactopentaosylceramide, 46%; sialyl-lactosylceramide, 50%; N-glycolylneuraminyl-neolactotetraosylceramide, 36%; and GM1, 30%. The N-deacetylated glycosphingolipids were re-N-acetylated quantitatively (96%) with [1-14C]acetic anhydride to obtain [acetyl-14C]glycosphingolipids of high specific activity.
