In this article I review our current understanding of some important lipid biosynthetic reactions taking place in the endoplasmic reticulum (ER) and Golgi apparatus, and call attention to the fact that significant amounts of diacylglycerol (DA G) must be formed at the Golgi apparatus if sphingomyelin (SM) synthesis occurs there. Since DA G functions as an intracellular second messenger l, it seems necessary to keep its concentration within the cell low, and thus a mechanism for eliminating DA G formed at the Golgi apparatus is needed. I suggest several mechanisms by which this could occur, and highlight the implications of some of these mechanisms on membrane lipid traffic through the ER-Golgi complex.
