Ovarian grafts in rats have been used for an analysis of factors leading to the destruction and elimination of transplants from alien hosts. Five series of grafts were studied with the genetic differential between graft and host varying systematically from an autogenous, through sibling, intra-strain, and homogenous, to a heterogenous relationship. The effect of ovari-ectomy of the host was studied in each series as it affected regeneration and repair of the graft, infiltration of leukocytes and connective tissue, and survival in an alien environment. Autografts nearly always succeeded in both ovariectomized and non- ovariectomized hosts; heterografts (mouse ovaries) invariably retrogressed. In the remaining series, ovariectomy of the host consistently tended to accentuate regeneration and to delay or prevent the destruction of grafts in an alien environment. No sign of incompatibility was evident between graft and host until 5-7 days after operation, when many grafts were rapidly destroyed. In grafts showing active regeneration the onset of degenerative changes was delayed for weeks or months. Homografts of ovary between unrelated strains of rats in some instances survived in good condition for at least 3 mos. These data are consistent with the concept of acquired immunity as a basis for host antagonism. They do not support Loeb''s theory of organismal differentials. It is suggested that transplanted cells may survive in an alien environment despite the presence of immune bodies by means of a neutralization or inactivation of these substances.
