A new, automated flow injection analysis (FIA) method was developed for rapid and specific determination of .beta.-lactams, e.g., penicillins and cephalosporins. The enthalpies of enzymatic reactions were measured in continuous flowing streams by thermistors. Enzyme reactions were carried out on small solid bed reactor columns, whereby the selectivity for various analytes was exclusively dependent on the specificity of the chosen enzyme system. Accuracy of enzyme thermistor FIA (ET-FIA) was evaluated by comparison with high performance liquid chromatography. Statistically no significant difference was found between the 2 methods. The minimal amount of K-penicillin V detectable by ET-FIA was 10-6 g. Precision of the method was determined by 10 consecutive injections of the same sample; the relative standard deviation was .+-. 0.8%. Linearity was obtained in a concentration range of 0.1-10 mg/ml for penicillins and cephalosporins. Sample capacity was 40-45 samples/h; analysis time for 1 single sample was < 80 s.
