Materials and MethodsReagents. Porcine insulin was a product of Novo. Pepsin was obtained from E. Merck. DTNB, TPCK-trypsin, collagenase (type II), and albumin (Bovine, fraction V) were Sigma products. Acetyltyrosine ethyl ester was from Dongfong Biochemicals, Shanghai. DTT was from Serva, mercapto-ethanol from Fluka, and TFA from Pierce. Urea was a local product twice recrystallized according to Morangos and1 Abbreviations: ATEE, acetyltyrosine ethyl ester; CD, circular dichroism; DP, despentapeptide (B26-30); DO, desoctapeptide (B23-30); DTNB, 5, 5'-dithiobis (2-nitrobenzoicacid); DTT, dithiothreitol; EDTA, ethylenediaminetetraacetic acid; HPLC, high-performance liquid chro-matography; TFA, trifluoroacetic acid; TPCK, Ll-[(p-toluene-sulfonyl) amino]-2-phenylmethyl chloromethyl ketone; Tris-HCl, tris-(hydroxymethyl) aminomethane hydrochloride.
