A molecular phylogenetic analysis of the genus Donacia (Coleoptera, Chrysomelidae) in Japan, based on mitochondrial gene sequences

The phylogenetic relationships among the species of Dona cia in Japan were analyzed using sequence data of mitochondrial cytochrome oxydase subunit I (COl) and l6S rRNA (l6S) gene regions. Using these markers, the subgenus Donaciomima was found to be paraphyletic, from which a monophyletic group containing the subgenera Dona cia and Cyphogaster was derived. This result is consistent with a previous morphological cladistic analysis. The sequence divergence between species is large, indicating relatively ancient speciation, except for a group with D. splendens splendens, D. splendens hiurai and D. tomin agai, which might have differentiated during the latter half of the Pleistocene. The divergence pattern in COl sequences indicates that D. tominagai and D. splendens hiurai are closer, also producing D. splendens paraphyletic species. The present analysis provides molecular phylogenetic evidence for Donacia yuasai being a synonym of D. provostii. The relationship between phylogeny and ecological characters is also discussed.
