The aminoglycoside antibiotics have been used for years to treat severe infections in infants and children. Their excellent in vitro activity against most Gram-negative Enterobacteriaceae and staphylococci is usually paralleled by favorable clinical results. The dis-covery of derivatives with anti-Pseudomonas activity and the introduction in clinical practice of amikacin, which is less susceptible to inactivation by bacterial enzymes, have in recent years' led to an improved spectrum of activity. On the other hand, it seems that no substantial improvement has been obtained as far the toxicologic aspects are concerned. In fact, the aminoglycosides used commonly still have important dose-related toxic effects that have been the object of intensive investigation.""
