The dauer larva of C. elegnas is a developmentally arrested stage formed at the 2nd molt under conditions of starvation or overcrowding. It is specialized for long-term survival and dispersal. Dauer-constitutive (daf) mutants form dauer larvae even in an environment favorable for growth. Temperature-sensitive (ts) mutants representing 6 different genes were subjected to temperature-shift and -pulse experiments in order to define the times of temperature sensitivity (TSP). The results for 5 of the mutants, including 1 mutant with a maternal effect, show TSP which bracket the 1st molt. The exceptional mutant, daf-14, has its TSP within the 1st larval stage. The TSP of 2 mutants, daf-8 and daf-14, exhibit distinct major and minor phases. Temperature-pulse experiments discriminate between alternate interpretation of the temperature-shift data. All the results can be interpreted on the basis of normal and defective execution stages for the ts functions at permissive and restrictive temperatures.
