MATERIALS AND METHODSSpecimens of the siphonophore Hippopodius hippopus were collected during spring in the bay of Villefranche-sur-mer (France), and gametes and cupules were obtained as described earlier [CarrC et al, 19811. Sizes of the cupules range 0.12 mm diameter. Polyacrylamide gradient gels [Neuhoff, 19731 were prepared inside microcapillaries (Intra-red Blaubrand) of 5 pl total volume (32 mm length and 0.4 mm diameter). Capillaries were half filled by suction of a solution of 0.35 M Tris-sulfate (Sigma, St. Louis) pH 8.8, 0.37% TEMED, and 0.75% ammonium persulfate (Biorad). Then, the second half of each capillary was filled by suction of a solution of 40% acrylamide (Serva), 0.8% bis-acrylamide, 0.35 M Tris-sulfate pH 8.8, and 0.37% TEMED. Electrophoreses were run for 45 min at 60 V with running buffers composed of 0.05 M Tris-base, 0.38 M glycine (Serva). The gels were extruded from
