Materials and methods. This method is a modification of a previously described synthesis of fluorescent nucleotides (5). The polysaccharide (5–100 mg/ml) in 0.1 M aqueous sodium borate, pH 8, was adjusted to 50% dimethyl sulfoxide (DMSO)(v/v). A stock solution of 10–20 mg/ml high-purity N-methylisatoic anhydride (MIA)(CAS 10328-92-4; Molecular Probes, Eugene, OR)(w/v) in neat DMSO was prepared by vortexing. This stock solution was used quickly or stored at 80 C for up to 72 h. The use of DMSO avoids the problem caused by the relative insolubility of MIA in water encountered in the previous synthesis (5). Different degrees of labeling are possible, but it is convenient to label at a ratio of one MANT fluorophore per
