Lytic infection of CV-1 cells with SV40 results in a sequence of ultrastructural alterations characterized by primary swelling of endoplasmic reticulum, followed by condensation of mitochondria, and finally, by swelling of most other membrane systems, including mitochondria and nuclear membranes. Lysosomes, stained for acid phosphatase activity or labeled with ferritin, were found to be relatively stable structures. They remained intact in cells which had become trypan blue stainable and in which the morphology of other organelles had become grossly distorted. Lytic SV40 infection was also characterized by an increase in the intracellular Na+/K+ratio. Important similarities between the pattern of cytopathogenesis resulting from SV40 infection and that resulting from a variety of nonviral causes are noted.
