Abstract A variety of selective radioiron probes have been engaged to study the mode of action of desferrioxamine in the rat. Four 59 Fe-labeled complexes were used, all of which were characterized by a high initial hepatic uptake. A quantitative radioautographic method for determining the cellular distribution of 59 Fe in rat liver demonstrated that two of the complexes (damaged RBC and precipitated ferritin) were selectively removed by the RE cells, whereas the remaining two complexes (soluble ferritin and hemoglobin bound to haptoglobin) labeled the parenchymal cell of the liver. DF was found to induce a marked increase in the whole body excretion of parenchymal labels, whereas the increase in excretion of RE cell probes was no greater than could be accounted for by iron recirculation to the parenchyma after release to transferrin. A higher proportion of radioiron was chelated from soluble ferritin 59 Fe than from hemoglobin 59 Fe, suggesting an interaction of the drug with parenchymal ferritin stores and not with iron released by heme catabolism. This was further supported by the close correlation between DF-induced excretion and the fraction of the different parenchymal radioiron probes assimilated into parenchymal ferritin stores.
