AimTo investigate the induction of osteogenic activity in bone marrow mesenchymal stem cells (BMSCs) by a nanofiber coagulation of KLD-12/recombinant human bone morphogenetic protein-2 (rhBMP-2) and establish a minimally invasive treatment plan for lumbar instability.Materials and MethodsIn vitro three-dimensional cultures of KLD-12/rhBMP-2 nanofiber gel (experimental group), rhBMP-2 (positive control), and KLD-12 (negative control) were used to induce osteogenic activity in BMSCs from New Zealand white rabbits. Proliferation and survival rates of BMSCs were detected using cell-counting kit 8 and calcein-AM/propidium iodide fluorescence staining, respectively. Bone calcium proteins and alkaline phosphatase (ALP) activity were detected using ELISA and ALP kit, respectively. Osteocalcin (OC) and collagen expression were determined using fluorescence-based real-time PCR and Western blot.ResultsWhile the proliferative activity of all three groups increased with culture time, that of the experimental and positive control groups were significantly higher than that of the negative control group at all times. Calcein-AM/PI fluorescence staining revealed many circular cells in the gel. The mean survival rate of the cells was 14 days, an increase by 92.26 ± 2.28%, 92.72 ± 2.4%, and 90.56 ± 3.5% for the experimental, positive, and negative control groups, respectively; intergroup differences were not statistically significant. The ALP activity, OC content, and mRNA and protein levels of OC and collagen type I were significantly higher in the experimental and positive control groups than in the negative control group.ConclusionKLD-12 allowed the slow release of rhBMP-2 to promote the bioactivity of BMSCs for improved osteoblast differentiation.
