In our recent publication (1), we report that the EGYEL motif, previously found to promote T22-reactivity, was depleted from the mature CD24 low pool of γδ-T cells that developed in H-2T–deficient mice, indicating that ligand was influencing the γδ-T cell receptor (TCR) repertoire (1). In her letter, Chien (2) suggests that this conclusion was invalid because, in reporting our analysis, we did not include the upstream tryptophan (W) residue in the motif, which she contends is critical for T22-reactivity (2). We have reanalyzed the data, which reveal that the W… EGYEL motif and all permutations thereof, are depleted from the CD24 low population that developed in H-2T–deficient mice (Fig. 1). This motif is found at background levels in tetramer− thymic γδ cells, as expected. Regarding the contention that the W residue is essential to T22-reactivity, while it was previously reported that a tryptophan to alanine replacement did substantially reduce binding to T22 in surface plasmon resonance analysis, this does not exclude the compatibility of other amino acids at this position (3).
