Cellular resistance to cytotoxic drugs is a major obstacle to cancer treatment. Resistant cancer cells can display a multidrug-resistant phenotype that involves simultaneous resistance to a spectrum of natural-product cytotoxic drugs. One form of multidrug resistance results from the expression of P-glycoprotein, the 170-kd protein product of the MDR1 (also known as PGY1) gene (7-5). P-glycoprotein, an adenosine triphosphate-binding cassette (ABC) transporter, functions as an efflux pump to decrease intracellular drug concentrations. Although P-glycoprotein expression appears to be associated with poor prognosis in some hematologic malignancies (4-7), associations with either chemosensitivity or clinical outcome have yet to be established for most solid tumors in adults. Recently, multidrug resistance-associated protein (MRP), another ABC transporter, was isolated from a drug resistant cell line (8). We (9) and other investigators (10) have reported transfection studies that have established that expression of MRP confers multidrug resistance. While there have been reports (11-15) on MRP expression in leukemias, neuroblastoma, and thyroid cancer, the expression pattern of MRP in commonly occurring solid cancers in adults has not been described.
