Regeneration of aortic endothelium in the rabbit was studied experimentally after removal of the endothelium from the lower abdominal aorta. The area initially denuded of endothelium is at 1st covered with platelets and leucocytes, but no large thrombus develops. Endothelium regenerates and gradually covers the greater part of the bared surface. However, even after 7 months, endothelium had still not completely covered the defect. New endothelial cells are formed by mitotic division from pre-existing endothelial cells. Despite previous reports to the contrary, we find no evidence that endothelium is formed from any other tissue or that there is anything unusual about the mode of nuclear division. The edge of the regenerating endothelium shows many giant-cells which resemble those described by previous workers in human and animal vessels.
