A 12% solution of ethanol-1-14C in normal saline was given by vein to 6 monkeys during the last 30 days of pregnancy; they were killed at 15 min, 90 min or 12 h and foetuses and placentae were removed and frozen. Sagittal sections were autoradiographed and unsectioned tissues were homogenized and assayed for14C. On the 6th or 15th day of pregnancy 2 groups of 28 hamsters were given a 50% solution of ethanol-14C in saline intraperitoneally and were killed and their tissues were assayed for14C. The fetal hamster had more unchanged ethanol than the maternal brain during the first 30 min after injection and fetal concentration of unchanged ethanol was higher in late pregnancy than before the placenta developed. Ethanol metabolism and excretion were slower in hamster and monkey foetuses than in the mothers and placenta and amniotic fluid had high ethanol contents. Higher concentrations of ethanol-14C and metabolites in the fetus were found in liver, pancreas, kidney, lung, thymus, myocardium and intestinal wall and the greatest uptake in the central nervous system was in the cerebellum.-P.
