Kinetics of amino acid incorporation have been studied in oocytes ofRana pipiensat all stages from the full-grown ovarian oocyte, through the period of pituitary-induced maturation and several hours beyond fertilization or artificial activation. Also, amino acid pool sizes in oocytes have been measured at all these stages. Pool sizes are subject to considerable natural variation in the eggs taken from different animals. However, the rate of flow of amino acid through the pool and, thus, the rate of protein synthesis, are relatively constant in eggs at the same stage, even when taken from different animals.The rate of protein synthesis is very low in the ovarian oocyte from an unstimulated female and in oocytes up to about 20 hours after pituitary stimulation. At this time the rate rises rapidly, reaches a maximum level of about 0.5 μg per oocyte per hour and remains essentially constant thereafter. Neither fertilization nor artificial activation have any observable quantitative effect on protein synthesis.
