The main intrinsic polypeptide (MIP, MW 26,500) of bovine lens fiber membranes was isolated and characterized. It was extracted from urea-treated lens membranes by chloroform-methanol. The extraction recovered 23% of the total membrane protein which is 1/3 of total MIP. Failure to extract MIP from buffer-washed membranes suggested that it was bound to matrix protein. Gas chromatography showed that it contained no sugar residues. On the basis of its amino acid composition, MIP had a low polarity and was different from the .beta.Bp crystallin chain and from other soluble crystallins. Immunochemically, it did not cross-react with any of the crystallins, which together with the other data indicated that MIP is a typical membrane component.
