Formulations (3) of dl-norgestrel were administered daily to groups of 3 women for 5 consecutive days. The serum levels of d-norgestrel were related to dosage of dl-norgestrel ingested. Peak concentrations in circulation of synthetic gestagen were attained a .5-3 h after oral administration, followed by a rapid and sharp decline in levels until the next dose. Three women received 500 .mu.g of dl-norgestrel and 50 .mu.g of ethinyl estradiol for 21 days followed by 6-7 days of no medication for 2 consecutive cycles. Gonadotropins remained suppressed for 4-6 days when therapy was discontinued. Daily concentrations of estradiol varied from less than 5-81 pg/ml, and there was no differnece in estrogen values during nontreatment and treatment days. Due to the long half life of norgestrel, the 1-wk pill-free interval is not long enough for complete recovery of the reproductive axis from inhibition of oral contraceptives.
