SV40-INDUCED EPENDYMOMAS IN NEWBORN HAMSTERS .1. VIRUS-TUMOR RELATIONSHIPS

Intracerebral inoculation of newborn hamsters with SV40 resulted in the formation of ependymomas. The latent period and frequency of tumor incidence depended on the virus concentration employed. Each of 12 animals injected with 10STCID5O of virus developed characteristic tumors, whereas 106TCID50 or less gave negative results during a 7 months'' period of observation. The latent period ranged from 87 to 137 days. No evidence of viral multiplication in the brain or other tissues of the hamster was found. Attempts to demonstrate virus in tumor extracts failed consistently. However, SV40 was found in trace amounts when intact tumor cells were seeded on monolayer cultures of African green monkey kidney cells. Only a small fraction of the tumor cells contained infectious material. No free virus was detectable in tissue-cultured tumor cells. None of the hamsters inoculated with a minimum of 105TCID50/ml of SV1, SV2, SV4, or SV5 on the day of birth showed any detectable effects during 7 months of observation.
