Upon irradiating adenovirion by ultraviolet light, viral DNA and a part of viral protein cosedimented faster than free DNA when examined in the presence of 4 M guanidine HC1 in sucrose gradient, indicating that protein became tightly bound to DNA. By sodium dodecylsulfate polyacrylamide gel electrophoresis, it was verified that the protein is cross-linked covalently to DNA and that polypeptide VII is the only polypeptide species present. Based on the available data on the size of nuclease digested fragments, a model of adenovirion chromatin consisting of a nucleosome-like subunit of polypeptide VII-hexamer was constructed.
