A study was conducted to determine potential direct molecular interactions between gamma1 laminin ofAnopheles gambiaeand circumsporozoite- and TRAP-related protein (CTRP) ofPlasmodium berghei. Gamma1 laminin was used as bait and the bait construct contained the V domain (i.e., the extracellular part of the protein that contains 4 EGF-type repeats). The prey contained only the TRP (thrombospondin) repeats of CTRP. The potential interaction between bait and prey was assessed using the yeast two-hybrid system, and the strength of interaction was evaluated by measuring β-galactosidase activity from liquid cultures. Results revealed that the use of CTRP prey constructs in combination with gamma1 laminin bait resulted in transcriptional activation. β-Galactosidase activity was observed from liquid cultures of the same yeast transformants. The TSP repeats in the CTRP construct activated transcription about ten-fold and the enzymatic activity was measured to be 191±9.5. The strong molecular interaction between theP. bergheiCTRP protein and the gamma1 subunit of theA. gambiaelaminin suggests an interactionin vivobetween the surface of the parasite and the basal lamina surrounding the midgut.
